Résumé
L’analyse phytochimique de l’extrait aqueux d’écorce de racine de Zanthoxylum zanthoxyloides (FAZ) [Lam.] Waterman (Rutaceae) révèle la présence de stérols et polyterpènes, de polyphénols, de flavonoïdes, de quinones ainsi que d’alcaloïdes. La richesse de cet extrait en composés phytochimiques est compatible avec les nombreux usages traditionnels de cette plante médicinale. L’étude toxicologique de FAZ a permis de déterminer par la méthode graphique de Miller et Tainter et par la méthode par calcul de Dragsted et Lang respectivement une dose létale 50% (DL50) égale à 4168±462mg/kg de poids corporel (PC) et 5500±875mg/kg de PC. Ces valeurs indiquent que cet extrait aqueux d’origine naturelle n’est pas toxique. Aux doses comprises entre 2,7 × 10−4 et 5,5 × 10−1 g/kg de PC, cet extrait aqueux induit une hypotension soutenue, dose-dépendante semblable à celle induite par l’acétylcholine (Ach) de 5,6 × 10−7 g/kg de PC à 5,5 × 10−4 g/kg de PC. Il réduit fortement l’hypertension induite par l’adrénaline (Adr) à 2,5 × 10−5 g/kg de PC. Ces résultats indiquent que l’extrait aqueux d’écorce de Zanthoxylum zanthoxyloides est un hypotenseur. Ils justifient l’utilisation traditionnelle de cette plante pour traiter l’hypertension artérielle. L’inhibition partielle de l’effet hypotensif de FAZ par l’atropine, antagoniste compétitif des récepteurs cholinergiques de type muscarinique, suggère la présence de deux types de principes actifs dans cet extrait aqueux: 1) les principes actifs cholinomimétiques de type muscarinique; 2) et les principes actifs non cholinomimétiques.
Abstract
The aqueous extract from Zanthoxylum zanthoxyloides (FAZ) [Lam.] Waterman (Rutaceae) contained sterols and polyterpens, polyphenols, flavonoids, quinon and alkaloids. The richness of this extract into phytochimic components is compatible with the full traditional use of this medicinal plant. The toxicological study of FAZ has permitted to determine respectively by Miller and Tainter’s method and Dragsted and Land’s method a LD50 of 4,168±462mg/kg of body weight (b.w) and 5,500±875mg/kg b.w. These values showed that this natural extract is not toxic. For doses ranging from 2.7 × 10−4 and 5.5 × 10−1 g/kg b.w, this aqueous extract induced a dose-dependent sustained hypotension similar to that induced by acetylcholine 5.6 × 10−7 g/kg b.w to 5.5 × 10−4 g/kg b.w. It reduced the hypertension induced by adrenalin at 2.5 × 10−5 g/kg b.w. These results showed that the aqueous extract from FAZ is a hypotensive substance. They justified the traditional use of this plant in the management of arterial hypertension. The partial inhibition of hypotension effect induced by FAZ caused by atropine, competitive antagonist of muscarinic cholinomimetic receptor, suggested the presence of two types of actives components in the aqueous extract: 1) muscarinic and cholinomimetic activity; 2) and non cholinomimetic activity.
