Zusammenfassung
Hintergrund
In den letzten Jahren wurden durch molekulare Tumortherapeutika wesentliche Fortschritte in der Behandlung maligner Erkrankungen erzielt. Hintergrund ist ein spezifischerer Angriff auf maligne Zellen unter Vermeidung von Begleittoxizität konventioneller antineoplastischer Therapie. Ein wichtiger Angriffspunkt heutiger molekularer Tumortherapie sind Kinasen, die durch kleine Kinaseinhibitoren oder – im Falle von Rezeptorkinasen – auch durch monoklonale Antikörper gehemmt werden können.
Material und Methoden
Diese Arbeit basiert auf einer Literaturrecherche in der Datenbank Pubmed sowie einschlägigen Lehrbüchern zum Thema der molekularen Onkologie und Kinaseinhibition.
Schlussfolgerung
Durch zielgerichteten Einsatz von kinaseinhibierenden Substanzen können verschiedene Krebserkrankungen selbst in fortgeschrittenen Stadien heutzutage besser therapiert werden. Dennoch kommt es auch hier häufig zu Therapieresistenz. Kritisch anzumerken ist zudem der hohe wirtschaftliche Preis dieser Substanzen.
Abstract
Background
In recent years molecular tumor therapy has dramatically improved the treatment of various types of cancer. Molecular therapy is expected to attack malignant cells more specifically with less side effects than classical chemotherapy. Kinases are key targets of current molecular cancer therapy. Both small molecules and monoclonal antibodies are used for kinase inhibition.
Material and methods
This article is based on a literature search of molecular cancer therapy and kinase inhibition in the PubMed library as well as modern textbooks of molecular oncology.
Conclusion
Using specific kinase inhibitors several types of tumors can now be successfully treated even in advanced stages; however, therapy resistance is a common problem. The dramatic treatment costs are also a critical point but kinase inhibitors are an important novel treatment strategy in oncology.
