Zusammenfassung

Unerwünschte Arzneimittelwirkungen (UAW) sind für die Arzneimitteltherapie bei Erwachsenen und Kindern von großer Bedeutung. Die allgemeine Aufmerksamkeit für die begrenzt verfügbare Evidenz einer sicheren und effektiven Arzneimitteltherapie in der Pädiatrie hat in den letzten Jahren stark zugenommen. Dies zeigt sich nicht zuletzt an der wachsenden Zahl europäischer und internationaler Initiativen, die Pharmakovigilanz bei Kindern zu verbessern. Der vorliegende Beitrag vermittelt einen Überblick über die derzeit verfügbaren Daten zur Epidemiologie von UAW bei Kindern allgemein und speziell im Bezug auf den Einsatz von nicht zugelassenen Medikamenten. Aktuelle Zahlen aus Deutschland werden vorgestellt und im Zusammenhang mit der internationalen Literatur diskutiert. Darüber hinaus werden verschiedene Methoden zum Erkennen und Erfassen von UAW und deren Potenzial für die Zukunft diskutiert.

Abstract

Adverse drug reactions (ADRs) are an important issue in both paediatric and adult drug therapy. In the past decade, awareness about the poor level of evidence regarding drug efficacy, safety, and effectiveness in children has increased substantially. This is reflected by the increasing number of European and international initiatives to improve paediatric pharmacovigilance. This report provides an overview of the current data on the epidemiology of ADRs in children in general and of those related to unlicensed and off-label drug use. Recent data from Germany are introduced and related to the international literature. Furthermore, various methods for ADR detection and their potential for future activities are discussed.
