Abstract

ZNF191 is a new zinc finger gene which has a 1107 bp open reading frame (ORF) and encodes a 368 amino acid protein including a putative DNA-binding domain of four Cys2His2 zinc finger motifs at its C-terminal region. The ZNF191 clDNA is located in the chromosome 18q12.1 region where it is known that a variety of hereditary diseases are related to. Probably, this protein has potential function of stimulating the gene transcription.

In order to examine the function and structure of ZNF191 protein, the ORF of ZNF191 and its zinc finger regionZNF191(243–368) genes were inserted into PTSA-18 expression vector by PCR amplification, then the constructed genes were expressed in the PTSA-18/B121 (DE3) system. The two proteins were purified by DEAE-52, CM-23 and Heparin-Sepharose 4B columns. The pooled proteins showed a single band as assayed by Coomasie Brillian Blue Staining of an SDS/polyacryamide gel.
