The possibility of obtaining seeds following placental pollinationin vitro

Možnost získání semen po placentálním opyleníin vitro

Abstract

The paper aims at verifying the possibility of obtaining seeds following placental pollination in more than 20 species of dicotyledons and monocotyledons. Excised placentae with ovules were cultivated in solid nutrient media in Petri dishes. Pollen was transferred from anthers onto placentae, its germinating capacity being enhanced by a drop of a 5 per cent sucrose in boric acid in certain cases. Fertilization of ovules resulting in the formation of viable seeds was obtained withNicotiana tabacum, Melandrium album andAgrostemma githago. Although withPaeonia officinalis, Viola tricolor, Antirrhinum majus, Galanthus nivalis, Narcissus pseudonarcissus, Clivia miniata andTulipa gesneriana normal seed set occurred, the seeds did not germinate even after being stimulated with GA3. The results indicate that the findings may be used for further embryological studies. The species are listed in which no seeds were obtained.

Abstract

V práci je ověřována možnost získání semen po placentálním oplození u více než 20 druhů dvojděložných a jednoděložných rostlin. Isolované placenty s vajíčky byly kultivovány na ztuženém živném mediu v Petriho miskách. Pyl byl přenášen v prašnících na placenty a jeho klíčivost byla někdy stimulována kapkou 5% sacharosy s kyselinou boritou. Oplození vajíček a klíčivých semen bylo dosaženo uNicotiana tabacum cv. ‘Wisconsin 38’,Papaver rhoeas, Papaver somniferum, Melandrium album aAgrostemma githago. UPaeonia officinalis, Viola tricolor, Antirrhinum majus, Galanthus nivalis, Narcissus pseudonarcissus, Clivia miniata aTulipa gesneriana byla získána normální semena, která však neklíčila ani po stimulaci GA3. Výsledky naznačují využití poznatků při dalších embryologických studiích. Druhy, u nichž nebyla získána semena, jsou uvedeny.
