Density of osteoid tissue and osteones at different degree of calcification

Abstract

Two direct technical approaches have been developed in order to establish the density of three types of isolated microscopic structures of bone tissue. The average density of material from osteoid tissue has been found to be 1.46 by employing the isopicnic centrifugation technique in a CsCl-water gradient. The density of cylindrical osteones has been calculated from the ratio between their weight and volume. Osteones at initial and final stage of calcification have a density of 2.005±0.018 and 2.211±0.018, respectively.

Résumé

Les densités de trois structures ostéoniques (liseré préosseux, ostéon au stade initial de calcification, ostéon complètement calcifié) ont été déterminées au moyen de techniques directes sur des spécimens obtenus par microdissection. On a pu ainsi établir que la densité moyenne du liseré préosseux mesurée par centrifugation isopicnique en gradient de densité est de 1,46. D'autre part le calcul des densités de spécimens cylindriques appliquant le rapport poids/volume a donné les résultats suivants: ostéons au stade initial de calcification 2,005±0,018, ostéons complètement calcifiés 2,211±0,018.

Zusammenfassung

Die jeweilige Dichte von drei osteonischen Strukturen (kalkfreie Knochenvorstufe, Osteone im Anfangsstadium der Verkalkung und vollständig verkalkte Osteone) ist durch eine direkte Technik an durch Mikrosektion gewonenen Proben bestimmt worden. Somit konnte festgestellt werden, daß die durchschnittliche Dichte der kalkfreien Vorstufe, die mittels isopyknischen Zentrifugierens in Dichtegradiente gemessen wurde, 1,46 beträgt. Andererseits hat die Berechnung der Dichte der Osteone unter Bezugnahme auf das Verhältnis Gewicht/Volumen, die auf zylinderförmige Proben angewandt wurde, folgende Resultate ergeben: Osteone im Anfangsstadium der Verkalkung 2,005±0,018, Osteone bei vollständiger Verkalkung 2,211±0,018.
