Zusammenfassung
Zur Ergänzung lichtoptischer Befunde bei experimenteller Maul- und Klauenseuche-Myokarditis wurden Mäuseherzen im Frühstadium, also am 1. und 2. Tage nach der Virusinfektion ultramikroskopisch untersucht. Am 1. Tage waren trotz deutlicher Virusvermehrung histologisch keine, ultramikroskopisch relativ unbedeutende Befunde (beginnendes Faserödem, leichte Verfettung) feststellbar. Am 2. Tage post inf. mit hohem Virusgehalt der Herzmuskulatur waren die ultramikroskopischen Befunde entsprechend den schon lichtoptisch starken Veränderungen deutlich ausgeprägt. Die histologisch verfetteten Fasern zeigen elektronenoptisch außer einer starken Fettanschoppung ein ausgeprägtes Ödem. Da ein Teil der Mitochondrien und dementsprechend die Myofibrillen intakt bleiben, dürfte es sich um eine reversible Schädigung handeln.
Die histologisch als Koagulationsnekrosen imponierenden Fasern stellen ultramikroskopisch Kolliquationsnekrosen dar, die als Folge eines massiven Ödems durch Auflösung sämtlicher Mitochondrien und Myofibrillen gekennzeichnet sind. Es wird aus den Befunden geschlossen, daß die intracelluläre Ödembildung das primäre pathogenetische Prinzip bei der Infektion mit dem MKS-Virus darzustellen scheint, nicht nur bei der Bildung von Virusbläschen im Epithel der Haut und Schleimhaut, sondern auch bei der Entstehung der Fasernekrosen in der Herzmuskulatur.
Summary
As a supplement to studies with the light microscope of experimental foot and mouth disease the hearts of mice were examined electron-microscopically during the early phases; that is, on the 1st and 2nd days after the virus infection. On the 1st day, despite an obvious increase in the virus, relative insignificant changes were made out electron-microscopically (beginning edema of the muscle fibers, mild fatty change). On the 2nd day with a heavy viral infection of the myocardium the submicroscopic studies showed severe changes corresponding with those seen with the light microscope.
Besides the histologically evident fatty change of the muscle fibers, there was electronmicroscopically an engorgement with fat and a severe edema. Since a portion of the mitochondria and accordingly the myofibrils remained intact, the changes probably represent a reversible damage.
The fibers seeming to have undergone coagulative necrosis histologically gave electronmicroscopically the appearance of liquefactive necrosis. This was the result of a massive edema and was characterized by dissolution of all mitochondria and myofibrils. From the results it has been concluded that the intracellular edema appears to represent the primary pathogenetic principal in the infection with the foot and mouth disease virus. This held true not only for the formation of vesicles in the epithelium of the skin and mucosa, but also for the development of the necrosis in the myocardial fibers.
