Carcinogene Wirkung von Azoäthan und Azoxyäthan an Ratten

Zusammenfassung

Azoäthan erzeugte nach subcutaner Injektion von wöchentlich 50 bzw. 100 mg pro kg Körpergewicht bei allen 22 überlebenden Ratten Leukämien und maligne Tumoren in den verschiedensten Organen. Lokale Sarkome an der Injektionsstelle traten in keinem Falle auf. Beobachtet wurden: Maligne Geschwülste in Leber (6), Vormagen(2), Nasenhöhle (2), Mamma (4), Ovar (1), ferner Leukämien und Retikulosen (9), mesenteriale Sarkome (1), sowie Aesthesioneuroepitheliome bzw. Neuroblastome des Riechepithels (2) und Hirntumoren (2). Die mittleren Induktionszeiten waren 260 bzw. 220 Tage. Im Wahrscheinlichkeitsnetz wurden lineare Regressionen von hoher Charakteristik entsprechend einer Normalverteilung gefunden, in die sich alle Neoplasmen unabhängig von ihrer Lokalisation einordnen. Deshalb wird ein gemeinsamer Wirkungsmechanismus angenommen. Azoxyäthan erzeugte nach einmaliger subcutaner Injektion ein lokales Sarkom an der Injektionsstelle und Adenocarcinome im Darm. Die Beziehungen der Azo- und Azoxy-alkane zu Alkylnitrosaminen, -hydrazinen und -hydrazonen werden diskutiert, ebenso die zum Cycasin als natürlich vorkommendem Azoxyalkanol-Glucosid.

Summary

Azoethane, after subcutaneous injections in two dosage groups (50 and 100 mg/kg body weight) once a week, induced leukemias and malignant tumors in various organs in all 22 surviving BD strain rats. No local sarcomas were observed at the site of the injections. The malignant tumors were observed in (number of rats in brackets): the liver (6), the forestomach (2), the nasal cavity (2), the mammary glands (4), and the ovary (1). In addition, there were leukemias and/or reticulosarcomatosis (9), mesenteric sarcomas (1), esthesioneuroepitheliomes of the olfactory epithelium (2), and brain tumors (2). The medium induction times in the two dosage groups were 260 and 220 days respectively. In the probability range (time and dose response relationships), highly characteristic linear regressions were found corresponding to a normal distribution (for all tumors observed), into which all tumors may be grouped independent of their localizations. Therefore, it is assumed that a similar mechanism of carcinogenic action occurred in all cases. Single subcutaneous injections of 250 mg of azoxyethane per kg body weight induced a local sarcoma at the site of injection and adenocarcinomas in the intestinal tract. The chemical relations of azo- and azoxyalkanes to alkylnitrosamines, alkyl-hydrazines, and to- hydrazones are discussed, as well as these to cycasin as a natural occurring azoxyalkane-glucosid.
