Summary
In guinea-pigs sensitized to dinitrochlorobenzene (DNCB) hyposensitization was studied by injections of different DN(C)B-conjugates, injections of the hapten (DNCB) and after x-radiation. The patch tests excised before and after hyposensitizing treatment were investigated histologically. — Results:

Subcutaneous injections of guinea-pig's serum-DNB-conjugates inhibit—according to the findings of Landsteiner and Chase [21]-immediate, but not delayed reactions. The contact reactions partly show a strong granulodiapedesis and granuloclasia, symptoms which are interpreted as results of preceding perivascular antigen-antibody-reactions.

Extracts of guinea-pig's epidermis-DNB-conjugates injected subcutaneously and intraperitoneally, reduce the patch test reactions at a higher degree than equivalent serum-DNB-conjugates, particularly in epicutaneously sensitized DNCB-animals. Even high doses of epidermal DNB-conjugates do not totally inhibit the reaction.

Intravenous injections of high doses of DNCB dissolved in tween 80 or contained in erythrocytes reduce or inhibit patch test reactivity regularly, but only temporarily.

Subsequent to whole body irradiation the degree of contact dermatitis reactions in guinea-pigs decreases to a minimum in parallel with the number of lymphocytes in the blood. The patch test reactions resemble at this stage macroscopically and histologically urticarial reactions. They were interpreted as reactions to simultaneously present antibodies.
Zusammenfassung
An 2,4-Dinitrochlorbenzol (DNCB)-kontaktallergischen Meerschweinchen (MS) wurden Desensibilisierungsversuche mit unterschiedlichen DNCB-Konjugaten, freiem Hapten sowie mittels Röntgen-Ganzbestrahlung durchgeführt. Die vor und unmittelbar nach der jeweiligen Desensibilisierung sowie späterhin applizierten Epicutantests wurden histologisch untersucht. Ergebnisse:

Subcutane Injektionen von MS-Serum-DNB-Konjugaten unterdrücken — in Übereinstimmung mit Landsteiner u. Chase [19] — anaphylaktische Früh-AgAkR, nicht aber Epicutanreaktionen vom Spättyp. Histologisch zum Teil verstärkte Granulodiapedese und- klasie letzterer werden als Folge einer durch perivasculäre Antigen-Reagin-Reaktionen hervorgerufenen Gefäßirritation gedeutet.

Subcutan sowie intraperitoneal injizierte Extrakte von MS-Epidermis-DNB-Konjugaten inhibieren die Epicutantest-Reaktionen stärker als Serum-DNB-Konjugate, insbesondere bei epicutan subtoxisch sensibilisierten MS, jedoch allenfalls subtotal und durchwegs nur passager.

Intravenöse Injektionen hoher DNCB-Dosen, gelöst in Tween 80 oder in Erythrocyten, bewirken starke bis völlige, ebenfalls passagere Reduzierung nachfolgender Kontaktreaktionen.

Bei röntgen-bestrahlten Tieren sinkt der Kontaktreaktionsgrad nahezu parallel mit der Leuko- bzw. Lymphocytenzahl im Blut auf ein Minimum. Die in dieser Phase makroskopisch und mikroskopisch urticariellen Testreaktionen werden als Ausdruck einer Reaktion mit konkomitierenden serumständigen Ak gedeutet.
