Summary
The distribution of demethylchlortetracycline between the intra-and extracellular space of rat livers has been measured in vivo and in vitro.
Demethylchlortetracycline (DMCT) is accumulated in liver cells against a concentration gradient. The concentration in bile is higher than in plasma.
The intracellular accumulation and the biliary excretion are diminished by application of probenecid (Benemid).
A saturation of the cellular uptake and biliary excretion has been demonstrated in nephrectomized animals and in normal rats during constant infusion of the antibiotic.
Intracellular accumulation has been shown to depend upon the temperature of incubation medium. The uptake of DMCT by liver slices is much higher at 37°C than at 0°C.
The concentration gradient of demethylchlortetracycline is not lowered by 2-,4-dinitrophenol.
The mechanism of the intracellular accumulation is discussed.
