Abstract
Most hybridomas derived from fusion of myeloma cells with immunized or hyperimmune spleen cells do not secrete antibody of the expected specificity. This is related to the fact that only a small fraction of the spleen cells which are able to fuse with the myeloma cells secrete specific antibodies (1). The fraction of specific PFC in a spleen immunized with SRBC is of the order of 0.1%. By appropriate in vitro immunization this low fraction can be increased by one to two orders of magnitude. This enrichment is due to both selective survival of antigen stimulated cells in culture and escape from in vivo regulatory mechanisms.
