A paracrine FGF, human FGF16, is 98.5% identical at the amino acid level to rodent Fgf16 (18). In humans, FGF16 deficiency is linked to a metacarpal 4–5 fusion defect with no other overt symptoms (19–22).

In contrast, FGF-16 is expressed at low levels in the heart during mouse development [23,32]. FGF-16 production is restricted to the endocardium and epicardium in the embryonic heart, and expression of both FGF-16 and FGF-20 has been linked to Wnt signaling including in cardiac development [16,26].
